A comparative study on fungal laccases immobilized on chitosan BABT

The phenoloxidase enzyme laccase from the cultures of the Pleurotus ostreatus and Botryosphaeria sp. and a commercial laccase from Aspergillus sp. were immobilized on chitosan of pharmaceutical degree by adsorption followed by crosslinking. Different immobilization conditions in relation to the granulometry of support and amount of enzymatic laccase extract used were tested, aiming at reaching high enzymatic activity with the immobilized enzyme. Two different substrates, ABTS and DMP, were used for the determination of enzymatic activity. The highest enzymatic activity was obtained when 1.0mg/mL of the enzymatic laccase extract from Botryosphaeria sp. was used with 1.0g of support (200 mesh). En la producción de hongos comestibles se utilizan diversos residuos agroindustriales como la paja de cebada, trigo y avena. Como resultado del cultivo, se producen grandes cantidades de sustrato residual, el cual puede potencialmente ser usado para procesos de biorremediación in situ. En el presente estudio, se cultivó el hongo comestible Pleurotus ostreatus (CP-50) en presencia del hidrocarburo aromático policíclico, antraceno. La cepa se cultivó en tres condiciones: 1) Agar con Extracto de Malta (EMA), CP-50 y antraceno (tratamiento); 2) EMA y CP-50 (testigo) y 3) EMA y antraceno (blanco). Effects of pH, Temperature and Agitation on the Decolourisation of Dyes by Laccase-Containing Enzyme Preparation from Pleurotus sajor-caju BABT

(1) Background: In this study, the effects of different pH values ​​(2.4, 3.2, 4.4 and 5.0), temperatures (30, 35, 40, 45 and 50°C) and agitation (100 rpm) on the enzymatic decolourisation of twenty-two dyes belonging to the chromophore groups anthraquinone, azo and triphenylmethane were assessed. (2) Methods: In all conditions, it was used a crude enzyme broth containing 30 U mL-1 laccases produced by Pleurotus sajor-caju PS-2001 in submerged process. (3) Results: Regarding the effects of pH values, the best results were obtained at pH 3.2 and 30°C, in which bleaching was observed for all dyes evaluated. Influence of initial pH of the growing medium on the activity, production and genes expression profiles of laccase of Pleurotus ostreatus in submerged fermentation Electron. J. Biotechnol.

Background: Enzymatic activity and laccase isoenzymes number of Pleurotus ostreatus grown in different pH values of the growing medium in submerged fermentation and incubated in buffer solutions of different initial pH values were determined. The expression profiles of five laccase genes (Lacc1, Lacc4, Lacc6, Lacc9 and Lacc10) in these cultures were also studied. Results: The highest laccases activity was obtained in cultures grown at initial pH of 4.5 and the lowest in cultures grown at initial pH of 8.5. Isoenzyme profiles were different in all the cases. Influence of iron and copper on the activity of laccases in Fusarium oxysporum f. sp. lycopersici BJM

ABSTRACT Fusarium oxysporum f. sp. lycopersici is a phytopathogenic fungus that causes vascular wilt in tomato plants. In this work we analyze the influence of metal salts such as iron and copper sulphate, as well as that of bathophenanthrolinedisulfonic acid (iron chelator) and bathocuproinedisulfonic acid (copper chelator) on the activity of laccases in the intra (IF) and extracellular fractions (EF) of the wild-type and the non-pathogenic mutant strain (rho1::hyg) of F. oxysporum. The results show that laccase activity in the IF fraction of the wild and mutant strain increased with the addition of iron chelator (53.4 and 114.32%; respectively). Mycoremediation of Congo red dye by filamentous fungi BJM

Azo, anthroquinone and triphenylmethane dyes are the major classes of synthetic colourants, which are difficult to degrade and have received considerable attention. Congo red, a diazo dye, is considered as a xenobiotic compound, and is recalcitrant to biodegradative processes. Nevertheless, during the last few years it has been demonstrated that several fungi, under certain environmental conditions, are able to transfer azo dyes to non toxic products using laccases. The aim of this work was to study the factors influencing mycoremediation of Congo red. Several basidiomycetes and deuteromycetes species were tested for the decolourisation of Congo red (0.05 g/l) in a semi synthetic broth at static and shaking conditions. Production of phenol-oxidases and peroxidases by fungi isolated from irrigated rice BJM

The aim of this work was to study the potential of fungus strains considered as prospective degraders for the herbicides quinclorac and propanil, for ligninolytic enzyme production. Eight fungal strains were grown in King's B liquid culture medium supplemented with 0.05% Remazol Brilliant Blue R (RBBR) and in liquid culture medium containing wheat bran as substrate. The enzymatic system assessment were: lignin peroxidase, manganese peroxidase and laccases. Results indicated differential patterns of ligninolytic enzyme production; the highest enzymatic activities were related to the production of lignin peroxidase. Among the strains, two (P3SA1F and P11SA2F) showed RBBR discoloration, suggesting the possibility of their application in bioremediation studies.
